That doesn't make even the slightest bit of sense. Kaido can sense haki, and therefore there is no "assuming" going on. His question was in the form of being shocked at Zoro using CoC.

Confirmed that the attack on Kaido was a CoC infused attack, as Enma literally absorbs and disperses haki, and Zoro has been leaking it.
No one can argue against that fact about Enma.
