Yes, Kisame respects the strength of the Sannin. This doesn’t make the statement valid for anything but an excuse—Kisame is willing to fight anyone in his DB break down, Kisamehas his strength compared to Jinchuuriki and bijuu and goes after a character much stronger than Jiraiya later.

It’s hype that never pays off which is why most of the community agrees it’s a convenient excuse Itachi uses to avoid escalating the situation. His actual mission there is to scare Danzo, he’s already accomplished it.
