Jack initially embodied a cruelty that people expected from Kaido. Instead, Kaido is just a brute that doesn't really kill. And Jack's personality gets watered down in Wano. From the feared Drought, to Jack the Drag-down.

Killer is a stage removed from a typical SMILE user. He failed the luck chance and is basically a Pleasure. But if Chopper were to ever make a drug, it'd be more likely be one that turns people like Killer back to normal, rather than awakening non existent animal traits.
