Nika and Joy Boy are mostly likely the same person, as I sincerely doubt "Joy Boy" is his real name, and Nika was stated to have freed the slaves and bring smiles to peoples faces. If Kaido believes someone can become Joy Boy, then it's more of a title, rather than a persons name, and bringing smiles to peoples faces fits the description of "Joy" boy.

Nika was his name. Joy Boy was his title. Luffy will be Joy Boy.

Huh, stuck out to me that King brought up "Joy Boy" and "Pirate King" in the same conversation in Chapter 1036, as if both are connected. Going by that, then maybe Luffy could be Joy Boy once he gets to Laugh Tale and becomes Pirate King.
