Well technically Vice captain means the pirate taking the lead the moment the captain misses for any reason and that's usually the strongest folk. At least that's how it works in the op verse.
People who take Zoro as vice captain are basically assuming that in a hypothetical case where Luffy didn't have a den den mushi on Dressrosa, Sanji would have to ask ''vice-captain Zoro'' for permission to attack the Big Mom ship.

How about in the sea train when Sanji was asking permission to act ahead of the crew. Who was the two giving him orders ? Luffy and zoro with luffy ultimately winning Sanji will never give zoro orders
