Yeah, Susano´s can be penetrated/broken into. The only issue is that Itachi has a special shield for his called the Yata Mirror, which doesn´t seem like you can break through it(though it´s never really explained in canon)

I mean, the implication does seem to be that they´re like super special hax weapons(the mirror and the blade he has), I don´t think it would necessarily be impossible for it to block Doffys' awakening
