I think Zoro should be familiar with the smell of blood and there's more than enough of it to give off a smell. But kicking him isn't going to determine if he's faking or not since he can literally fake being in pain. It's a scene meant to show Zoros level headedness but it's just not written well.

Well, Zoro can sense the level of Haki. If he let Loki go, it’s because he knew Loki isn’t a big deal, at least not for Luffy. Luffy will probably just play around and smile during the fight. Then, someone will tell Loki that Luffy is Joy Boy, and the fight will end right there because Loki is just a fanboy of Joy Boy.
