The Marines are not known for being over-confident. They are always talking in terms of bringing overwhelming numbers to face opponents. Kizaru himself said that they would need to make "special preparations" to capture a version of Rayleigh who had been retired for 20 years.

This same Kizaru who showed overwhelming amounts of cautiousness against Rayleigh was like "2 Yonko? I can handle that. What you think Sakazuki?" And Sakazuki was like "Now, hold tf on Kizaru. Sending you to face 2 Yonko?...I mean that's fine, but we don't know how strong the Samurai are!"

Keep in mind, Sakazuki did not say that the Samurai were too strong for Kizaru, but rather that their strength was unknown Which also confirms that the strength of the Yonko was in fact known and that the Marines know 1 Admiral > 2 Yonko.

Zoro's endurance and stamina didn't drop or decrease over the time skip. It's required a combination of factors for him to fall which includes but not limited to eating poisonous food, starving, serious injury, and more.
