I think only the "chosen ones" being able to counter the Holy Knights would be extremely lame and run counter to Oda's style.
It seems much more likely to me the counter is a matter of knowledge and technique, not special bloodlines and secret abilities. This isn't Gaban's first encounter with an HK, and there's also really no reason to believe it's only his second. We know so little of what the Roger Pirates got up to or when.

Personally I think the D blood line is special in some way & given they are said to be the enemy of god, there’s gotta be a reason why. Even Imu has once said that D’s have forgotten what they are actually capable of or sumin along those lines iirc but I digress.
Knowledge & Technique will also probably be a counter to it after all Gaban is not a D as far as we know & Roger never seemed to possess the Nika fruit. I just think expecting everyone including those that don’t even know how to use Haki to learn a special Haki technique in such a short time is unrealistic. It also somewhat cheapens what regen is actually supposed to be (mid hats individually would actually stand a chance against Marco for instance with this 💀). I mean even durability monsters like Kaido or Linlin required extreme finesse or hax in order to be damaged but we can disagree on this 🫂.
