Yes.-Only the last loss counts, unless you think his losses to Crocodile, Lucci, Katakuri, Caesar, Doflamingo, weren't overcome by his eventual victory, in which case you are delusional.

Also outside of combat, for the war to be PK Kaido has already taken many big setbacks from a result of Luffy's alliance:
He has already lost SMILE, and Doflamingo to Luffy's fights, two massive blows to him.
If Kaido loses this fight, he loses control of Wano and completely loses potential to become pirate king.
Luffy has lost nothing material to Kaido at the moment, and has only gained in combat strength dramatically since reaching the island.
